Yes, after you finish your 50 pages you'll jump and read your favorite genre. Your next roll will be from the favorite genre square. Hopefully this helps and have fun!

Roll 1: I got a 1, so 50 pages. Doing this between 2 books I'm reading right now, Blood of Elves (the witcher) and Winter's Heart (wheel of time). For the go ahead do I also have to do the favorite genre challenge? I was a little confused by that.
