Castle Roogna in Piers Anthony's Xanth series has a giant, intelligent spider who's one of the main characters.

There is also a "classic" Dr. Who book about spiders Doctor Who and the Planet of the Spiders

Fund this while looking for the click for the Dr. Who book, so no idea if it's good or no. Lord Of The Spiders Or Blades Of Mars

The Master Mind of Mars The title "master minds" are a very weird race that's kind of spiderish but kind of not. Still, it's a cracking good story. (And if you aren't aware of it, Edgar Rice Burroughs is also the creator of TARZAN.)

Oh, and if you don't mind vaguely crab-like beings (I've always thought that crabs looked a LOT like spiders) there is Their Majesties' Bucketeers Another oldie but goodie.

I'm just reading this and was gonna post it. Good think is you don't need to read part 1 to read this book.

Also the aliens in Blindsight are kind of spider-like but they are more alien then spidery.

And of course The Hobbit and The Lord of the Rings have gigantic evil spiders (who have important episodes, but aren't actual characters).

Anansi Boys, adult novel about the contemporary children of Anansi, the African spider god of stories.

Ann Bishop's Black Jewels series, of adult and explicitly dark-fantasy works, features a race of sentient spiders as tertiary characters and a lot weaving and web imagery. The spiders specifically appear in the original trilogy and the Dreams Made Flesh collection.
